Say I wanted to sample a sound from WinAmp in another audio application.

I tried setting Winamp's DirectSound config to the Digital out, which then sends to the spdif output.

I then have a digital rca cable from the out of the spdif back to the input, but I don't ever get a signal on the spdif input in my control panel.

Will this work?

S/PDIF transfers data between devices. The source device must provide the clock. You don't have a source device to provide the clock.

I don't know your particular soundcardm but many external soundcards do not have any way to handle internal computer audio (unless, of course, it is to send it to some other external device or other computer). When direct recording of this sort can be handled by the soundcard, it is done by virtue of mixer panel settings. Many internal (PCI or built-in) soundcards can do this, but not all, as it is not considered a proper task for professional audio.

Lacking hardware to accomplish this task, you must use a speciality audio application. One of these is called Total Recorder

Another is Virtual Audio Cables http://spider.nrcde.ru/music/software/eng/vac.html
Others exist.
