Line in

I play my guitar through the line in input on my PC, I cant see how to utilise fruityloop 5 to process the signal for real-time playing. FL 5 ignores the signal and I cant find where to change the input settings, or whatever I need to adjust, to make this happen. Can anybody please help. Many thanks
